An abnormal growth mutant in maize has a defective mitochondrial cytochrome oxidase gene.

چکیده
We describe a new maternally inherited maize mutation, nonchromosomal stripe 5 (NCS5), that adversely affects plant growth and yield. Mutant plants are characterized by reduced height, defective yellow striping on leaves, and aborted kernels on ears. NCS5 striped plants carry both normal and partially deleted versions of the mitochondrial cytochrome oxidase subunit 2 gene and exhibit greatly reduced levels of cox2 transcripts when compared with nonstriped control plants. Other mitochondrial genes and their mRNAs are not affected. Thus, the defective plant phenotype is correlated with a reduction in the number of functional cytochrome oxidase subunit 2 genes. The NCS5 mutant mitochondrial genome appears to have arisen by amplification of a rare homologous recombination product.

Differential expression of alternative oxidase genes in maize mitochondrial mutants.
We have examined the expression of three alternative oxidase (aox) genes in two types of maize mitochondrial mutants. Nonchromosomal stripe (NCS) mutants carry mitochondrial DNA deletions that affect subunits of respiratory complexes and show constitutively defective growth. Cytoplasmic male-sterile (CMS) mutants have mitochondrial DNA rearrangements, but they are impaired for mitochondrial fun...
متن کاملAnalysis of Leaf Sectors in the NCS6 Mitochondrial Mutant of Maize.
The nonchromosomal stripe (NCS6) mutation of maize is a partial deletion of the mitochondrial cytochrome oxidase subunit 2 (Cox2) gene. The Cox2 deletion and a narrow yellow striping phenotype are inherited together in a maternal fashion. The striped plants are heteroplasmic for mutant and normal Cox2 genes. Only the mutant Cox2 gene is detected within the yellow stripes, whereas both normal an...
متن کاملPPR2263, a DYW-Subgroup Pentatricopeptide repeat protein, is required for mitochondrial nad5 and cob transcript editing, mitochondrion biogenesis, and maize growth.
RNA editing plays an important role in organelle gene expression in various organisms, including flowering plants, changing the nucleotide information at precise sites. Here, we present evidence that the maize (Zea mays) nuclear gene Pentatricopeptide repeat 2263 (PPR2263) encoding a DYW domain-containing PPR protein is required for RNA editing in the mitochondrial NADH dehydrogenase5 (nad5) an...
متن کاملAssociation of Pathogenic Missense and Nonsense Mutations in Mitochondrial COII Gene with Familial Adenomatous Polyposis (FAP)
Nuclear genetic mutations have been extensively investigated in solid tumors. However, the role of the mitochondrial genome remains uncertain. Since the metabolism of solid tumors is associated with aerobic glycolysis and high lactate production, tumors may have mitochondrial dysfunctions. Familial adenomatous polyposis (FAP) is a rare form of colorectal cancer and an autosomal dominant inheri...
متن کاملEvaluation of effectiveness of some mitochondrial genes in biosystematics and phylogeographic studies of house mouse (Mus musculus ) subspecies
The identification of the efficiency of some mtDNA genes of Mus musculus species complex (house mouse) for biosystematics research was studied in this approach. Recent studies have made use of different mitochondrial genes including NADH dehydrogenase genes, cytochrome b gene, cytochrome oxidase genes, D-loop region and whole mtDNA genome to study the house mouse species. Usage of each of these...
